The Transformation Of Bacteria ( Green Fluorescent Protein )

Transformation in bacteria is something that could be essential for survival in a bacteria. In order to perform this transformation naturally a bacterium must considered competent, otherwise it must undergo an artificial transformation. Being a competent cell means that the bacteria can take up DNA from its environment naturally (5). Those that are not competent such as Escherichia coli that are not naturally competent can be tested with an artificial transformation, such as what we will use in this experiment. Methods used can obtain things such as chemical mutagens or radiation (1). The gene used for the transfer is the GFP gene (Green Fluorescent Protein), which gives an illuminating appearance under a UV light when conducted properly,†¦show more content†¦Experiments introduce the idea that the uptake of DNA by competent bacteria can be the result for survival reasons such as need for food or evolution purposes (5). The difference between natural and artificial gene tra nsformation in a bacteria, however, with those who are incompetent versus competent allow he or she to conduct an experiment with artificial transformation. By doing this one may be able to determine and observe the formation of biofilms or mutations by transferring a gene to another organism for beneficial or harmful purpose. This could lead to finding resistants or if a gene introduced has any effect on the bacteria. In the experiment, the pGLO plasmid contained encodes the gene for GFP as well as a gene for resistance to an antibiotic allowing a transformation to take place when adding a carbohydrate such as arabinose to the medium. Research has been found on the horizontal gene transfer on how virulence factors are acquired as well as genes resistant to antibiotics spread with microorganisms (5). By conducting a similar experiment he or she can observe similar findings or observe the results and conduct some sort of understanding of these ideas. The findings in this experiment a re to show a positive result in what causes growth and the glowing when the plasmid pGLO is added to the positive as well as the difference in the medium. Robotics Essay Example For Students

Robotics Essay Two years ago, the Chrysler corporation completely gutted its Windsor, Ontario, car assembly plant and within six weeks had installed an entirely new factory inside the building. It was a marvel of engineering. When it came time to go to work, a whole new work force marched onto the assembly line. There on opening day was a crew of 150 industrial robots. Industrial robots dont look anything like the androids from sci-fi books and movies. They dont act like the evil Daleks or a fusspot C-3P0. If anything, the industrial robots toiling on the Chrysler line resemble elegant swans or baby brontosauruses with their fat, squat bodies, long arched necks and small heads. An industrial robot is essentially a long manipulator arm that holds tools such as welding guns or motorized screwdrivers or grippers for picking up objects. The robots working at Chrysler and in numerous other modern factories are extremely adept at performing highly specialized tasks one robot may spray paint car parts wh ile another does spots welds while another pours radioactive chemicals. Robots are ideal workers: they never get bored and they work around the clock. Whats even more important, theyre flexible. By altering its programming you can instruct a robot to take on different tasks. This is largely what sets robots apart from other machines; try as you might you cant make your washing machine do the dishes. Although some critics complain that robots are stealing much-needed jobs away from people, so far theyve been given only the dreariest, dirtiest, most soul-destroying work. The word robot is Slav in origin and is related to the words for work and worker. Robots first appeared in a play, Rossums Universal Robots, written in 1920 by the Czech playwright, Karel Capek. The play tells of an engineer who designs man-like machines that have no human weakness and become immensely popular. However, when the robots are used for war they rebel against their human masters. Though industrial robots d o dull, dehumanizing work, they are nevertheless a delight to watch as they crane their long necks, swivel their heads and poke about the area where they work. They satisfy that vague longing to see the human body reflected in a machine, to see a living function translated into mechanical parts, as one writer has said. Just as much fun are the numerous personal robots now on the market, the most popular of which is HERO, manufactured by Heathkit. Looking like a plastic step-stool on wheels, HERO can lift objects with its one clawed arm and utter computer-synthesized speech. Theres Hubot, too, which comes with a television screen face, flashing lights and a computer keyboard that pulls out from its stomach. Hubot moves at a pace of 30 cm per second and can function as a burglar alarm and a wake up service. Several years ago, the swank department store Neiman-Marcus sold a robot pet, named Wires. When you boil all the feathers out of the hype, HERO, Hubot, Wires et. al. are really jus t super toys. You may dream of living like a slothful sultan surrounded by a coterie of metal maids, but any further automation in your home will instead include things like lights that switch on automatically when the natural light dims or carpets with permanent suction systems built into them. One of the earliest attempts at a robot design was a machine, nicknamed Shakey by its inventor because it was so wobbly on its feet. Today, poor Shakey is a rusting pile of metal sitting in the corner of a California laboratory. Robot engineers have since realized that the greater challenge is not in putting together the nuts and bolts, but rather in devising the lists of instructions the software that tell robots what to do.Software has indeed become increasingly sophisticated year by year. The Canadian weather service now employs a program called METEO which translates weather reports from English to French. There are computer programs that diagnose medical ailments and locate valuable o re deposits. Still other computer programs play and win at chess, checkers and go.As a results, robots are undoubtedly getting smarter. The Diffracto company in Windsor is one of the worlds leading designers and makers of machine vision. A robot outfitted with Diffracto eyes can find a part, distinguish it from another part and even examine it for flaws. Diffracto is now working on a tomato sorter which examines colour, looking for no-red i.e.unripe tomatoes as they roll past its TV camera eye. When an unripe tomato is spotted, a computer directs a robot arm to pick out the pale fruit.Another Diffracto system helps the space shuttles Canadarm pick up satellites from space. This sensor looks for reflections on a satellites gleaming surface and can determine the position and speed of the satellite as it whirls through the sky. It tells the astronaut when the satellite is in the right position to be snatched up by the space arm.The biggest challenge in robotics today is making softwa re that can help robots find their way around a complex and chaotic world. Seemingly sophisticated tasks such as robots do in the factories can often be relatively easy to program, while the ordinary, everyday things people do walking, reading a letter, planning a trip to the grocery store turn out to be incredibly difficult. The day has still to come when a computer program can do anything more than a highly specialized and very orderly task. The trouble with having a robot in the house for example, is that life there is so unpredictable, as it is everywhere else outside the assembly line. In a house, chairs get moved around, there is invariably some clutter on the floor, kids and pets are always running around. Robots work efficiently on the assembly line where there is no variation, but they are not good at improvisation. Robots are disco, not jazz. The irony in having a robot housekeeper is that you would have to keep your house perfectly tidy with every item in the same place all the time so that your metal maid could get around. Many of the computer scientists who are attempting to make robots brighter are said to working in the field of Artificial Intelligence, or AI. These researchers face a huge dilemma because there is no real consensus as to what intelligence is. Many in AI hold the view that the human mind works according to a set of formal rules. They believe that the mind is a clockwork mechanism and that human judgement is simply calculation. Once these formal rules of thought can be discovered, they will simply be applied to machines. Thais Review Paper Essay on Thais The reader should cherish the book. Do not put it down reversal, not to bend pages, not to carry it without the package in a dirty bag. Thats probably the few rules that hammered into my head. All this set is called concern about the book. And sometimes you have so that the book is a concern for you? This happened with Thais. From the first lines of the preface and explanatory notes I was warm and comfortable, as if covered soft knitted blanket. Here is an explanation of the measures of length and weight, here are the names of months. However, even if the explanations were not, it would not be a problem. The interpretation of the values of well-woven into the text. Here and there, one of the characters asks casually, and clarifies something where the value is shown in parentheses. Before we assess the artistic content of the book, we should not forget that this is something like a historical reconstruction. It is like to collect old clay pot of hundreds of fragments it is difficult, long and not always harmoniously. And Thais , there are fragments, where the action is somewhat sketchy, but Efremov cleverly takes the other. In any product there is a hanging fragments. Some people, the meeting which was, however, not vpletetsya then into the main fabric of the narrative, the place to which the hero never return. In terms of depiction Ephraim ravnyh- no clothes fabric and slide under the fingers, face breathes fresh Cretan breeze the nose is filled with the dust of Egypt. Everything is alive, sharp and realistic. Thin characters noble men and elegant women complete the picture. Sometimes, I myself would like to put the book down and walk easily and gracefully as Greek hetero, holding high his head and shooting eyes at passers. index a good book for me is the desire to meet the hero. Without looking up, I read about an excellent education, how humbly and with all the patience Thais immersed in the religious mysteries of the different people, how calm she conducts interviews with famous people and how caring about slaves and those who serve it. A lot of all of the above is not applied to our reality, of course, but wanted to read more, sit a little straighter, gentle smile and be a little bit better than it actually is The most exciting in the book -. It is just tight tangle of religious teachings. Shedding light on the mysteries of the cult of the Mother Mother, Orphic, the Stoics and others, Thais know how many similar deities and their images have evolved over time and the improvement of the human mind and a knowledge. Efremov said at the beginning that set himself the task to walk on the time of formation of a new religious attitude and the birth of humanism, and especially pleased that the conductor of this world become intelligent and stunningly beautiful woman.

What are the benefits of emergency management doctrine, and how do you Essay - 3

What are the benefits of emergency management doctrine, and how do you think its creation may be enhanced in the context of the UAE - Essay Example ces had to evacuate their homes for the sake of visitors and holy servants who crowded the area for such functions and since the climatic conditions are always harsh, the inhabitants had to move around all day long distributing water to quench thirst. The United Arab Emirates, for the last eighty years has been on the move coming up with an effective emergency management plan. From the Saudi Arabian documentation, the first response to such emergencies was by the establishment of a fire brigade in 1927 to serve pilgrims who flock Makah each year for special prayers. The establishment of emergency management doctrine has brought along great benefits to the United Arab Emirates. It has been possible to save lives and assets from damage. United Arabs Emirates has been the centre of discussion because of the terrorist attacks for example the Khobar tower bomb attack in 1996, which claimed 19 lives, the Yanbu and Asir floods caused pure destruction to property estimated over 230,000 km2, the fire and meningitis outbreak during Hajj and Ramadan festivals in 1997 and 1987 respectively affected close to 2,000 pilgrims (Alamri 15).. These are just a few examples of the damages caused by hazardous happenings. Through emergency management doctrine, it has been able to contain most of the calamities through resource optimization; which means being in a position to deploy and respond to emergency situations. For instance, due to a daily increase in the number of worshippers flocking Makkah, the Saudi Red Crescent Society has already come up with a medical e vacuation system which involves the use of twenty five helicopters. This in turn will help save innocent lives in case of disease outbreaks. In addition, it has eased the motor vehicle crashes and traffic jams. This is evident through the creation of Makah Metro, which has the capability to connect all the holy places around Makah 24/7. This works in line with a digital traffic control system; Saher. This ensures all

Flight to Mexico - General Aviation - Essay Example The â€Å"pilot to be† must pass the FAA practice flight test. In addition, a private pilot must be able to speak, read and comprehend English. A medical examination for aviation is also vital and can only be administered by a medical official certified by the aviation department. It is critical that a private pilot has all the necessary documents. These documents consist of photo identification, a student pilot certificate with endorsements, a FAA Form 8710-1 Airman Certificate. Moreover, a private pilot needs to have a logbook with CFI endorsements along with along with an aircraft maintenance logbook. As a private pilot, tourist VISA is extremely essential along with FMM, which is a visitor permit that is provided by the airline. In addition, a customs declaration form is vital. Clearing immigration upon arrival is highly suggested for foreign pilots since clerks do check these documents. For further approval, the FMM permit gets stamped by the airport officials. As mentioned above, strict guidelines are placed in all aspects of transportation, which includes customs. Since Mexican customs deal with a lot of traveling and shipping options, they are strict in not permitting certain things to enter their soil. One of the main objectives of Mexican customs is to ensure the protection of their country. Hence, it is quite common for the Mexican government to reject goods that seems susipicious and is not labeled clearly. Everything must be cleared with US customs since both customers understand the gravity of security issues that emerged post September 11. Any monetary value over ten thousand dollars must be approved and declared with the customs. Mexico is a beautiful country that has the ability to spark excitement and awe amongst tourists. Mexico offers beautiful beaches along with historical cities that date all the way to Spanish colony. Mexico City itself offers many incentives such as great

The Importance of Being Earnest Essay Example for Free

The Importance of Being Earnest Essay The comedy in Importance of Being Earnest focuses on the aristocratic classes at the turn of the 19th century, the time when the play was written and set. Whilst creating humour by mocking their absurdities i. e. moral and social values and ideas, he also provides the audience the witty banter of the idle rich and their somewhat ridiculous lifestyles and attitudes to enjoy. These idle rich seem to revel in making trivial matters important and important matters trivial. Wilde uses a range of techniques to simultaneously mock aristocratic Victorian society and to create humour: dramatic irony; the dialogue and language of the characters i. e. puns/wordplay, paradoxes and their witticisms; the significance of the themes at the time of the play and today; the actions of the characters; characterisation and the development of the increasingly ridiculous plot, which all culminates in the discovery that Jack was the baby in the handbag. The play begins in a realistic atmosphere for the time that the play was set; the characters behave and talk in the leisurely, pointed and conscious manner of their day. The bachelors only relax when they are alone. We see their idle nature almost immediately through the characters trivial preoccupation with food, one of the themes of the play. In the first few lines of dialogue in the opening scene, we witness Wildes witty way with words and the consequent humour he creates in the scene. Algernon, avoids admitting that he plays poorly and claims that he plays with wonderful expression, as though doing so were more transcendent. He emerges to be quite a conceited character and it seems that Wilde intends him to represent the pompous nature of the rich, rather like Lady Bracknell does. Algernons dialogue is always polished and his quips, paradoxes and unorthodox attitudes provide quite a lot of the humour in the play. Sometimes, what the characters say might sound intelligent but in fact, is silly. It is hidden by the sophisticated sentence structures. Wildes use of language is rarely simple: as Gwendolen says, it is style not sincerity which is important, i. e. he cares more for a statement being clever rather than true. The characters in The Importance Of Being Earnest are all language conscious. The sentences they speak are perfectly formed and a reflection of how aristocrats would like to speak, not how they did speak at the time. They all try to sound intelligent. At all times, even at tense times, the characters prioritise the upholding of a fake mask of politeness. An example of this is when Lady Bracknell says rise from this semi-recumbent posture at once. Again it is a mockery of Victorian aristocrats. Both Dr Chasuble, who is somewhat pedantic, and Miss Prism show an explicit interest in the language they use, they repeatedly state where their metaphors are drawn from: Were I fortunate enough to be Miss Prisms pupil, I would hang upon her lips. I spoke metaphorically my metaphor was drawn from bee.

Dune by Frank Herbert is one of the greatest Science Fiction novels of all time. The story is about Paul Atreids as he tries to take back his planet (Arrakis, a.k.a Dune a pure desert world. It is also the only place where spice can be found. Communism and Democratic Capitalism

1. From a Christian perspective, why did Marxist Communism fail? The government has too much power of constrains people. Trying to force people and form them to change is not right and does not work in the long run. To give any one party too much power is a bad idea. We are all human and prone to sin. The government needs to help the people and work for the people not rule them. 2. Which is a more Christian form of government, democratic capitalism or democratic socialism? That is a tricky question, and I believe the answer lies somewhere in between the two.The struggle is to regulate but not over regulate the people. People are greedy and in capitalist economy will not give to the poor. That’s were socialism looks better, but then the government is to power full and human nature can affect the government just as it can a civilian. I believe my vote goes for democratic capitalism with less government and a competitive market. 3. What functions does government have to undertake because of fallen human nature? The government has to protect society from itself so to speak. The government becomes responsible for creating laws to protect rights and property. They also become responsible for enforcing these laws and protecting people from others who may steal their things. The government also has to be responsible to take care of the environment since people fail to do it on their own. Another thing I see the government doing is taking care of people who can’t devise the means to take care of themselves. 4. Should Christians concern themselves with the regulatory activities of government? If so, what areas of regulation are most significant?Yes I do believe Christians should concern themselves with regulatory activities. The major one would be discrimination. There needs to be very strict laws guarding against regulation. All of god’s children are created equal and all should be treated fairly and given the basic tenants of life. Also everyone abl e should be able to work and have to opportunity to support themselves and contribute to society. Another regulation that needs to be of concern to Christians is over regulation. Over regulation can lead to many deceitful and bad things when abused by greedy individuals and government.
